Post subject: Residual acrylamide in wells
|
Anyone have the issue of residual acrylamide left in wells after pulling the comb from a gel? It's as if the tolerance of the comb thickness is not that well controlled. Any tips as to how to eliminate this (aside from buying the precast gels)?

Hi,
make sure that you use dry combs. I often got that problem, if i use wet combs.
In addition we polymerize our gels at 37°C.
Give it a try.

You can try rinsing your wells with water after removing the comb...this is what we do in our lab and it seems to work!

we too rinse wells with buffer (and use dry combs) and do not experience this problem

1. Use a dry comb and also dry and clean plates;
2. Take a syringe with a needle and fill it with buffer-,
3. Insert the needle tip in side a well (do not puncture it) and wash by pressing the syringe plunger multiple times. Press well so u get a "jet" out of the needle.U will see unpolymerized AA swimming out the well into the buffer reservoir.
4. Repeat multiple times washing untill u see no AA swimming out;
